阿爾瑪藍(Alamar Blue)是一種細胞增殖檢測試劑,對各種人和哺乳動物細胞、細菌和真菌提供一種快速和敏感的細胞增殖和毒性檢測方法。
阿爾瑪藍(Alamar Blue)是一種基于刃天青(resazurin)的檢測試劑。刃天青作為一種一種氧化還原(REDOX)指示劑,根據細胞代謝還原情況發生顏色變化。氧化態的刃天青呈紫藍色且基本無熒光,其還原態產物試鹵靈(resorufin)轉變為粉紅色且高度熒光,產生的熒光強度與呼吸活細胞數量呈正比。通過檢測呼吸過程中氧化水平,阿爾瑪藍用作一種定量檢測細胞活力和毒性的直接指示劑。阿爾瑪藍的顏色變化可通過普通分光光度計檢測吸光度變化,檢測波長為570nm,參考波長為600nm;阿爾瑪藍的熒光變化可通過熒光光度計檢測,激發波長在530~560nm之間,發射波長為590nm。
檢測原理:(細胞增殖實驗 Cell proliferation assay))
※ 生長中細胞引起Alamar Blue的化學還原反應;
※ 持續性細胞生長維持還原環境,使得Alamar Blue呈紅色,發強熒光;
※ 生長受抑制維持氧化環境,使得Alamar Blue呈藍色,無熒光;
※ 使用基于熒光或吸光值檢測的儀器收集數據;
※ 熒光檢測的激發波長為530~560nm之間,發射波長為590nm;
※ 吸光值檢測的波長為570nm,參考波長為600nm;
保存方法:
室溫12個月穩定,2-8°C保存20個月穩定;避光保存;
